I am just trying to try out tubeless tyres but...

Fitted a pair of Scott Cougar tubeless tyres to my XT wheels a couple of weeks back; really tight to fit and initial inflation a pain, but managed it with a track pump.
Went on holiday and left bike behind, came back and spotted I had fitted one tyre backwards, removed, refitted and added some sealant. Now I cannot achieve inflation, even after going to the local garage and using their car inflator!
What am i doing wrong?

did you clean the rim and the bead of the tyre before the refit?

maybe some dried sealeant on either causing it not to seal fully?

Make sure the tyre is nicely on the rim, then push down on the tyre (pushing the sidewalls against the rim) by the valve, then pump.

I did'nt clean before refit as bike had not been ridden and no sealant used first time.

I'll try pushing tyre down before another workout on the pump!

How long can the sealant be exposed to air before its knacked?

yup as real man says push down on the tyre over the valve so the bead seats there otherwise you never get it to inflate:

This might also help:
