Yes as long as the total capacity is ok, which, by what you describe will be.

Yes, I think alot of raod mechs specify a 27 tooth max on the rear but I have run a 105 and Tiagra with a 28 tooth SLX cassette and it works fine.

The 2010 Ultegra 6700 has a 11-28 cassette but you need the 6700 rear mech to run it with - allegedly.

ditto what 59 times the pain said, i'm running dura-ace mech on 11-28 slx cassette with xtr shifter just fine. ran tiagra before that and that worked fine too, all on 26t granny 34t middle and bash.

Not sure how relevant this is, but years ago I ran a 12-28 seven-speed cassette and a 24, 34, 46 chainset with a set of DX thumb shifters and a 105 rear mech. Never had a problem.

That was super cool back in the day! A 48 or a 52t chainring and an ultegra/Dura Ace rear mech.....
