    Is it possible to plot a road/off-road route on Google Earth and measure its distance? If so, how d'you do it?

    I just select the ruler tool and choose the path setting. You can then plot the route and get a distance. Sadly you can't then save the route however.

    google pedometer is the tool for mapping offroad routes I think

    Just use Mapmyride.com – same info plus a tool that maps, measures and stores routes – magic!

    Not quite as well integrated as mapmyride.com, but it has Ordnance Survey maps too. Much better for offroad route planning.
