Leaving Glasgow at roughly 6pm heading to the Institute of Sport in Sheffield in a minibus - what's the best route? Google suggests that there's not much in it.
Also, anything to do in Sheffield other than watch the BUCS indoors athletics that I'm taking folk to? Unfortunately no bike.
M74, M6, A66, A1, M18, M1 (North)- IMHO of course, but speaking as a Glaswegian ex-pat living in York.
You could of course take the M6 as far as Manchester and cut across one of the passes, but I personally wouldn't go that way as you will lose a lot of time in the Manchester area.

That's the way we go too, so definitely the best way.
Personally.... Scotch corner then south - but don't do the M18 bit, stick with the M1. The non motorway bit of the A1(M) towards the M18 is grim by comparison. I'd avoid going via manchester, it can add a lot of time to the journey

Road numbers will be about right - but it ain't quick round there.
Which side of Sheffield are you heading for - east or west?
Also if you take the A1 route, then don't take the M1 where the A1 and M1 split - stay on the A1 and take the M18.
Just noticed a 6pm set off, so scenic routes not high on the agenda. Manchester will be no problem other than rush hour, which it won't be
As for stuff to do, check out what's on at the plug, leadmill and octagon for live music. Decent clothes shops round division st area if you can handle arty types and students

At that time of day I'd do M74, M6, M61, M60, M621, M67 then either Snake (A57) or Woodhead (A628). I drive around Manchester/Sheff daily and used to live in Scotland trying to get to Holmfirth and that was always the best way. M621's usually a big mess but at that time it'll be clear.
M621? going via Leeds city centre? definitely not.

you could go and have a wander around 'shag town' (attercliffe), see if there's a 'special interest' club that takes your fancy?
(and there's a few awesome old tool shops to poke around - they're good for an hour or so)
I'm west of Glasgow (Bishopton), my own family in Sheffield & Barnsley. Given your start time, I'd go with the M74/M6/M61/M60 clockwise/M67/A57 Snake Pass combo, around 250 miles.
I worked at "Billy Smart's Circus" (British Steel Corporation) as a lad. If timing suits, you could visit Kelham Island (http://www.youtube.com/watch?v=3YfTtGCsiD8). If you have the space, take a bike, find Wharncliffe Woods and look out for Peaty or just head back out the A57 around Ladybower Res.
South 
For the EIS there is no point at all in going via Manchester as that brings you into Sheffield on completely the wrong side. I live on the west side of Sheffield and my parents are in Carlisle and even from there I've found it quicker and easier on the whole to go via Scotch Corner. Coming down via SC then off the M1 at J34 drops you only a minute or two from EIS so definitely the best option IMO.
matthew h makes a good point though most direct woodhead (A628) and more so the snake (A57) take you to wrong side of sheffield plus i wouldn't fancy being a passenger in a minibus on either
M62 has very long sections 50mph roadworks with speed cameras west of M1/M62 junction so i'd do A66 (assuming weather permits) Scotchcorner A1M to M1 J34

not my scene but Meadowhall shopping/cinemas 5 minutes drive
the on one / planet x shop is about 10 minutes drive from EIS
trams run every 10 mins or 20mins depending on time of day to city centre from near IES or as someone suggested Kelham Island Museum and Fat Cat pub and other real ale venues also can be done by tram
