Can't see why not if a) it doesn't interfere with getting it in your jig and b) you haven't got one of those wheels with the nipples at the hub end of the spokes.

Yes, you'd have to remove the cassette if its on the drive side. So you take it off just like any other wheel with a broken spoke.
