Much as I still love Afan/Glyncorrwg, I'm feeling the need for something new, but that will still provide enough challenging trails for a weekend away with friends. Where else is worth a trip up from the South East? I'd love to go to Scotland but probably not worth the drive for a weekend only!

Yup, half the distance to get there and plenty of riding to keep you more than happy for the weekend if like rocky trails.
Dalby forest was great for a weekend.
I've only ridden the Scotch ones which are pretty far for you. I think many would recommend the Tweed Valley for Inners/GT and if you've not ridden them then I think you'd enjoy them - red and black one gay at GT, cx and some DH at Inners the next.
Otherwise the other stanes are also good and varied with a few quite close to each other in the SW (of Scotland).

Len - Have you been riding in N Wales? Go and ride Coed y Brenin and Penmachno or Nant yr Arian if you definitely want trail centres.
Otherwise South Lakes has enough riding for a month, let alone a weekend, and doesn't take that long to reach.
Yeah, CyB & Penmachno definitely worth a trip.
Or as suggested, plenty of good riding in the Peaks, which isn't too far.
Assuming you can read a map Peaks & Lakes as above. For Lakes base yourself somewhere near Ambleside. There are many sources of route guides & maps inc all the bike shops in the Lakes who sell copies of all the classic routes for a few £s.

Why not go and base yourself for a weekend at Machynlleth?
That way you've got a choice. You could ride the Cli-machx or any of the Mach loops. Then you've got 2 trail centres only 30-40 mins away.
Coed-y-Brenin (enough here for a couple of days)
&
Nant-yr-Arian (if you're feeling epic you can even cycle to it, off piste, from Mach)
Loads of variety and enough 'wild' riding if you fancy some non trail centre stuff.
