Got to present to work colleagues top tips on preparing for and taking part in a charity 45 km bike ride, I understand they are mostly novice cyclists so want I need are top tips & pearls of wisdom for this audience??

Drink as much as you can during the ride
Ride as much as possible before the ride

Proper shorts and Assos chamois creme, ride as much as they can between now and then to break in their arses. Then ride some more.

I already had ...

Ride your bike a lot (esp. if you don't already)
Helmet
Proper shorts
Clean your bike - the bits that move especially
Know how to fix a puncture
Bring enough water and food

There must be some more pearls of wisdom out there ??

Make sure their saddles are the right height.

Nothing worse than watching people grinding pedals sat on a low seat with minimal leg extension.

Group 'practice' training rides to see where they are at fitness wise and for you to identify/sort out their bike set up problems.

slick tyres for mountain bikes presuming it's on road.
