Just redecorated spare room in new house, all walls painted successfully apart from one? Paint peeling off when rollered? Any ideas?

Contamination of some sort on the original painted surface. Could be anything, if it's a wall where the head end of a bed has been it could be hair grease/hair product residue, or it could be furniture polish if previously behind a dressing table etc. What prep did you do? If you can scrape it back, sand and wash with sugar soap you should be o.k.

I used a sander to smooth off any filling i'd done and get rid of any old paint.

That's your problem. You need to prime the filler before top coating. Ideally you need a contract matt with little or no vinyl in it to create a base coat. I usually use white contract matt on ceilings and use a bit of that to spot prime filler. Whats happening is that the new paint isn't soaking into the filled areas causing air pockets which then come away when you roller over them. You might get away with watering down the emulsion you have to mist coat the filled areas. Depends if it's a silk finish or an emulsion with loads of vinyl in it. Hope that helps
