Been running just QR's (Salsa) on my Simple for a couple of years now. Swapped the allen head bolts out for Hex and no problems so far. Live in South Wales so there's a fair bit of hammer going through the chain.

I ran a QR Rohloff in a horizontal dropout Chameleon, despite Rohloff recommending using the nutted version.
Some QRs were 100% reliable, some slipped at the first hill. I later progressed to an EBB, then a sliding dropout frame and can't now remember which were the good QRs.
If by Bolt Up, you mean the skewers with a socket cap head that are actually a very long thin bolt, I found them 100% reliable.
Never tried chain tugs.

I use a shimano quick release with a chaintug on each side, just to make sure. Haven't had any slippage so far.

Think I'll start with chaintugs and quick release.

I thought Simples had a little screw adjuster to hold the axle at its set point?

Using Bolts here just for the security and I've had QR move around-just me? others I know have used QR no problem too.

Had a qr on it's own, then went for allen bolts, now use allen bolts and chain tugs

tugs + bolt up always worked nicely on my bmx! My last bmx didn't actually have tugs but I find it a lot easier to align the wheel and keep it aligned if there are tugs. I suspect they'll be handy for keeping it in place with a disc brake as well.

binned the little screws as they are too little.

Chaintug on the drive side with a shimano QR.
i sometimes get movement on the disk side.

Wouldn't want to run fixed with just a QR in track ends mind. Anything with the potential to unship the chain could be lethal.

The screws are fine if you swap them out for hex heads.
