When it comes out of the tube it's a liquid but presumably dries to the paste/putty like consistency found on rotor bolts and other pre-treated threads.

To actually apply it should I thread the bolt with the threadlock still wet or wait for it to firm up first? Does it make any difference?

Just run round a couple of threads when wet and then screw it in. Dont let it set before you screw the bolt in.

I put it in wet. But there's no reason why you can't let it dry first - like on pre-treated bolts.

I was under the impression that Locktight .243 and similar threadlocks cured in the absence of air and hence you may be waiting a long time for it dry in the fresh air.

Yep, I'm sure the dry stuff you find on bolts is a different type.

Have spilt it before - took a long time to dry!

It will only cure under pressure and anaerobically. the paste on new bolts is different.
