Specifically the Wheelsmith stuff.
Do I need to remove the old stuff from spoke threads when relacing a wheel?
Will be using new nipples if that helps.
I'd say if you're concerned enough about a wheel build that you use spoke prep vs dab of grease etc, then you'd be better using new spokes.
However, IME old spokes don't need anything. You may find the old prep binds a little on a couple of spokes, but try a new nipple on one and see how much there is. Once you put tension on it it'll barely make a difference.
I may be more inclined to dab threadlock on the end of the spoke/nipple once built, depends on the spokes and how springy they are.
I expect they should be fine so long as the tension is nice and even? I'll crack on without removing it I think, new nips seem to meet a little resistance from the old stuff.
Cheers!
