I have now got parts to build up a singlespeed - does it need a fixed rear wheel or can you use qr hubs?

You can use a QR hub, thus it will have a freewheel. Or you can buy a dedicated singlespeed hub with a thread to take a srcre-on freewheel or a fixed cog.

i would recommend using a normal freehub wheel; allows you to change cogs and sort out chainline. also freewheels unless you spend a lot.
use a qr with a chaintug (assuming trackends) or bolt up without; doesn't make too much difference.
