The new Polka Dot Challenge (Spud Riley) for quiet roads: http://www.polkadotchallenge.co.uk/Polka-Dot-Route.asp

The old Polka Dot Challenge for the classic climbs: Cat and Fiddle, Buxton, Long Hill, Whaley Bridge, Glossop, Snake, Winnats, Axe Edge, Gun Hill

Do a search for the route of the Tour of the Peak, that should be about right as a starter.

My mate did 10 laps of Winnats in a day, no dabs on Winnats!
Hope, Castleton, Winnats, Mam Nick, Edale and round. Brutal.

Try this one:
http://www.peakepic.co.uk/p/routes.html

Route of a new Sportive in the Peak District, starts from Bakewell.

Tour of the Peak is another good suggestion, nice route that. Map here:

I do a variation on that route quite regularly, usually miss out the Holme Moss bit and add in some more stuff further south though.

Double strines be a good start, plenty of climbing on there.

If you come from ladybower end can ride down to midhopestones then up to Langsett and back over strines.
