I have just bought some hope mini IS mounted brakes on here and I need adaptors. I have IS fitting on forks and rear of bike. Running 160 front and 140 back.Do I just need a Hope Mount Adaptor IS to IS +20mm at the front,and the rear 140 bolts on directly, using shims to align?
160 and 140 should bolt straight to the fork and frame without adapters as long as it is all IS to IS.
It all depends on what calipers you have. For IS hope calipers, they do different models for different rotors.
The same model of caliper will fit a 20mm bigger rotor at the front than the back.

Then they should just bolt straight to the frame and fork and take a 160f and 140r rotor with no adaptors. Adding the adaptor you mention would allow you to fit a 20mm bigger rotor than currently (eg 180f, 160r)
If they're the older hope brakes which are direct IS mounts without an adaptor then the caliper number will determine what size rotor you need.

On the back of the caliper just above the lower mount hole - should be 5 for rear 140mm IS

Yes, that'll be fine - that Hope link refers to the older discs that were 5mm larger.
