Stupid question time - looks like I need to disconnect my Guide brake to route the cable correctly through the (full sus) frame
To minimise bleeding requirement is it better to disconnect at the caliper or lever end?

I always do it at the lever if possible, less chance of spilled fluid on the friction surfaces, and usually only needs the lever bleeding afterwards. If you can get your hands on a red barb from a reverb fitting kit, it'll minimize fluid loss from the hose too.
