Centred around Bakewell would be cool
Wow, what a difficult question. There is so much good road riding around there that provided you stay off the busy roads you won't go too far wrong. There's some quite tough climbs too. If it was me I'd get out of the valley and onto the tops west of Bakewell. Beeley Moor (south east of Bakewell) has some cracking views but the cars you do encounter tend to be tanking it. Riding through Chatsworth is always worth doing if you've not done it before.
My "commute" involved going from Staffordshire MOorlands to Bakewell and Beeley MOor. I've always fancied turning that into a circular road ride. There are stunning views and some serious climbs. Clues: Bakewell, Monyash, Hurdlow, Crowdecote, Longnor, up to Leek - Buxton road (good long climb), Morridge/Mermaid pub, Warslow, Hartington etc. That would be lovely. ๐
[EDIT] no idea how long that is but if your idea of 50 miles is relatively flat this will keep you busy I'm sure
That wouldn't be far off 50 miles I reckon, just make sure the brakes are in good working order for the drop down Crowdecote.
Come back from Hartington via Long Dale/Parsley Hay/Over Haddon and you'd have a cracking ride.
I'd head north - Bakewell, Ashford, Hassop, Calver, Froggatt, Fox House, Hathersage, Edale, Castleton, Tideswell, Litton, Gt Longstone, Baslow, Chatsworth, Bakewell.
Plenty of little roads to play on too.

Really, you won't go too far wrong. Both routes go in and out of white and dark peak.
Mine is from here (Hazel Grove'ish), Marple, Marple Bridge, Charlesworth, Glossop, Snake Pass, Bamford, Hathersage, Surprise View, Fox Inn, Calver, Bakewell, Buxton, Long Hill, Whaley Bridge, Disley, Hazel Grove(ish). You could cut through Chapel after coming down Long Hill and back over to Glossop via Hayfield/Little Hayfield and then chunnel to get shorten the mileage.
Only one snake on the A57 Spoonie, hence 'Snake Pass' ๐

Sheffield->Glossop via Manchester Road/Snakes Pass
Glossop (first left, past the golf club, then left again at the end)-> hayfied -> Chappel
Chappel (turn left, Sheffield Road, Winnats pass) -> Hope Valley -> Sheffield

Be careful between Hayfield and Chinley (before the left under the bridge to Chapel). A friend was very nearly killed instantly on this road. She died about 5 years later of complications caused by the crash. She's the reason they changed it to a 50 limit. I would never ride that road.

Only time I've ever had a problem on there was during a Sportive when loads of riders were on it and drivers got really pissed off. Snake Pass was closed for ages a couple of years ago due to landslides so it was great to ride then. We used to do it as part of night time road rides and there'd be loads of roadies out on it!
But everyone knows that the proper way to do Snake Pass is up from Bamford and down into Glossop.

road[/url]. Heading NE from the A6 towards Millers Dale. Fantastic descent and the climb out to Tideswell is nice too.
In fact Rob, I've been threatened with being shown how to ride a road bike properly by Crazy Legs for a long while. We could all three of us do a ride. A singletrackroad ride.

With respect, that's a bloody awful ride. You can dodge the whole hideous Hayfield to Chinley section with some cunning use of back lanes or bypass it altogether. I wouldn't touch that section of road, it's too narrow and too fast. Awful.
[edit - more helpfully, you can dive into Hayfield, ride up Highgate Lane, do a short section up to Peep of Day then take the back lane down into Chinley. Better still, use a cross bike and you won't ride even that sort section of road]
The Snake is okay early morning, but vile when busy on summer weekends, but there are so many quieter options around, go over the Strines Road for example and thread you way back via Holme Moss using mostly back lanes. The Woodhead's another particularly vile road btw.
Anyway...
Bakewell, Longcliffe, Ashbourne, Cheadle or Leek, Warslow, Longnor, Monyash.
You can do it all on minor (ish) roads.
