Can't guarantee it, but I did exactly that ion the Alps with 2.5in tyres (Too tight to buy new tubes for them) and I didn't get a puncture.
I regularly run 2.1 tubes in 2.35 tyres too, don't seem to get any more punctures than anyone else.

I've got a 2.1 Tesco tube in my 2.7 highroller just now 😳 That's the sort of rock solid race prep that earns you 33rd out of 34 baby 8)
