[Closed] using a spoke key - school me!

Got me a spoke key and got me a roadie wheel with a loose spoke that isn't quite true. The wheel's not massively out of true, still fits within the brake blocks without excessive rubbing, so I'm hoping to get away with sorting it myself.
Wheel building and fixing is a frontier I have yet to cross - how do I go about doing this?

And don't forget, anticlockwise to tighten(because effectively you work from the wrong side of the nipple,scratched my head for a while over that the first time.)
