I've always sized a new chain by wrapping it big ring front and rear and adding two links.
Is this any different now I have a roller underneath the chain tensioning it?

Yes, a bit. TBH the best thing to do is just test it- if it's a suss bike, unbolt the shock and swing the arm through the travel. big big +2 is only ever a bodge really.
