Anyhow, it is a fair drive from Cheltenham to the start and obviously it isn't a loop so i have to get from the end to the beginning.
What are the options, how late is the last train, or is does it have to be done on multiple days because of the transfer times?

I've completed the SDW in a day; you can do it in 11-12 hours. Best way is to stay overnight in the Youth Hostel in Winchester and make an early start. Eastbourne has regular train services to London & Brighton for getting home. There is an annual organised, sponsored ride. I'm doing it again this year 🙂

I did it over 2 days on the last bank holiday weekend on my cross bike - and will definately go to do it in a day later in summer. I think I'd be doing it all in a day - trains at both ends etc - to minimise the load on the back. Route finding is easy except if it's busy and you're being polite and mindful of others at intersections - then miss signs! Finding water never seemed to be a problem. Trains can get you to eastbourne quite early, and they run until reasonably late in the evening from winchester IIRC, especially if you're looking at 11-12 hours.
Thought I'd better add that it's easy starting from Winchester as you have the prevailing winds behind you.
