I've got 3 spacers with my new Hope BB, what's the optimum position for my triple crank running a 34t on the middle?

That is not what the spacers are for.
If you have a 68mm BB Shell you need two on the drive side and one on the non drive side. With a 73mm BB shell just one spacer drive side. If you need to change your chain line (which you won't)you can buy chain ring spacers (search ebay) as you have a triple all you need is singlespeed chainring bolts and to put your new single ring where the middle ring should be.
Easy.

let's say you were converting from a dedicated 2x10 crank to 1x10.... what size spacers would one need?
