Do you think a reasonable off-road route between Cardiff and London exists?

Yes.

The Ridgeway will take care of the middle bit.

Fill in with this http://www.bridlewaymap.com/ and some OS map scrutinising

Not all that handy, though, the bridleway map. Just clicked and remembered why I don't use it! Lots that's not on there. You'd give up on the project if you looked at that!

edit - less sure now. Depends how much cheeky you fancy, and too much footpath might slow you down quite a bit if you get into overgrown paths, stiles, kissing gates etc. Reckon you might have to settle for a chunk of Sustrans type back roads.

Getting to Bristol seems to only be possible on road, but then it's Bristol -> Bath cycleway, then the canal to Devizes, up onto the Ridgeway to Streatley, into Reading, over to Swinley then pick up the Thames path at Weybridge.

Not sure how I could get much of that good Hampshire riding in besides Swinley though.

Leave the K&A at Devizes, drop S towards Imber Perimeter Path on Salisbury Plain then keep heading E to Collingbourne Woods for singletrack fun. Alternatively follow the ridge which you can pick up just N of Devizes then follow all the way to Watership Down territory. Perhaps head for Basingstoke Canal?

If you gave Collingbourne Woods a miss then head for Combe Gibbet and follow Wayfarer's Walk all the way to Watership Down.

Cyclestreets.net always seems mad keen to send me on bridleways when I select quietest route.

There are two clear routes - ridgeway and CG's suggestion. Interesting.

You could do the wessex way from tim woodcock's book

https://www.amazon.co.uk/Wessex-Way-Minehead-Wheelwrights-Mountain/dp/1859810608

You could turn left at shoreham and go up the downs link, then follow the reverse of the offroad london to brighton
