I used it on my Lapierre Froggy with no problems when I had it. The downtube on it was not completely round and there were no issues.

The clamp on the 591 is quite big so most bikes should fit securely in it.

Have 2 591s, and they will clamp just about any shape as long as there isn’t a suspension linkage or somethign in the way. The jaws are V-shaped at the botton and flat on top, so a rectangly tube should be fine.

The Outride is a neat solution if in doubt

Does anyone have any experience of the Thule 591 bike carrier with the Giant Defy 1? The downtube is quite rectangular and before I buy the 591, I want to be confident that the clamp will securely grasp the downtube. I don’t want the bike coming loose and potentially damaging either the Giant or the car.

Thule have not been able to advise me.
